Enzyme Histochemical and Immunohistochemical Evaluation of Human Lymph Node in Plastic Section

Abstract
The present work was designed to evaluate the enzyme histochemical and immunohistochemical characteristics of various cells that constitute lymph node.
Lymph nodes were obtained at surgery for the evaluation of lymphadenopathy and were embedded in glycerol methacrylate(plastic) through the complexing tissue processing procedures. Enzyme histochemical examinations('5nucleotidase, adenosine
triphosphatase, acid phosphatase, alkaline phosphatase, nonspecific esterase, and peroxidase) were performed on thin sectioned(2§­) tissues. And immunohistochemical stains for IgG and S-100 protein were also performed.
@ES The results were as follows:
@EN 1. The plastic-embedded tissues showed signfiicant histologic improvement and precise correlation of enzymes with morphologic characteristics compared with frozen and paraffin sections.
@ES 2. Enzyme histochemical examinations:
@EN 1) Cells of mononuclear phagocyte system(MPS) including macrophages, sinus histiocytes, interdigitating reticulum cells(IRDCs) and follicular dendritic cells(FDCs) showed strong reactivity on 5'-nucleotidase, adenosine triphosphatase, and
nonspecific esterase stains.
2) Subgroups of lymphoid cells including T lymphocytes, mantle zone lymphocytes, and plasma cells could be identified specifically. Mantle zone lymphocytes showed positive reaction on 5'-nucleotidase, adenosine triphosphatase, and alkaline
triphosphatase staina. And T cells located on paracortex exhibited positivity on acid phosphatase and nonspecific esterase stains.
3) Vascular endothelial cells were weakly to moderately positive on 5'-nucleotidase, adenosine triphosphatase and alkaline phosphatase. And the endothelial cells of postcapillary venules(high endothelial venules) showed weakly positive reaction
on
acid
phosphatase and ¥á-naphthyl acetate esterzse, but these were not significant findings.
3. On immunohistochemical examination, FDSs is germinal center and IDRCs in paracortex showed strong cytoplasmic positive reaction on S-100 protein stain. And some lymphoblasts in germinal center and plasma cells in medullary cord showed
positivity on
IgG stain.
According to these results, simultaneous applieation of enzyme histochemical and immunohistochemical examinations on plastic embedded lymphoid tissue will be useful in identifying T lymphocytes, B lymphocytes and cells of mononuclear phagocyte
system.
It also will be extremely useful in the precise diagnosis and classification of the neoplasms derived from these cells.
